NanoPro 1000 XDR Assays
ProteinSimple's new NanoPro 1000 XDR reagents and protocols offer advanced protein signal enhancement capabilities and a broader dynamic range of detection, while also providing more protocol flexibility on the NanoPro 1000 system.

NanoPro 1000 XDR assays are designed for use on the NanoPro 1000 system, and must be used in conjunction with NanPro 1000 XDR Master and Amplified Secondary Antibody Detection Kits, Compass Software version 1.8 or higher and compatible firmware. NanoPro Peroxide XDR
Peroxide XDR expands the upper dynamic range of detection for NanoPro 1000 assays, enabling detection of proteins when using higher concentrations of lysates. This is achieved with an improved peroxide formulation that produces a more robust signal with minimal substrate depletion (signal burn-out).

Dynamic Range of Detection for ERK1 in HeLa Lysate. A comparison of substrate preparations using Peroxide/Luminol and Peroxide XDR/Luminol was tested with increasing amounts of HeLa lysate ERK1 protein levels. The new Peroxide XDR extended the upper dynamic range of ERK1 detection by 2X.

Detection of High Protein Concentration with Minimal Substrate Depletion. MCF10A lysate at 0.2 mg/mL was probed with an anti-Hsp70 primary antibody. The profile using Peroxide/Luminol substrate (top) shows no distinct signal detected due to substrate depletion (signal burn-out) caused by the high amount of lysate used in the assay. In contrast, a distinct Hsp70 profile is observed with Peroxide XDR/Luminol substrate (bottom).
Amplified Secondary Antibody Detection Kits
Amplified Secondary Antibody Detection Kits enhance sensitivity at low protein concentrations. Amplification binding via streptavidin-biotin interaction is utilized to increase signal intensity, resulting in a lower limit of protein detection for NanoPro 1000 assays.

Multi-Step Incubation Protocols
The new NanoPro 1000 protocols provide more flexibility in assay configuration, allowing up to three incubation steps per assay. Designed for use with Peroxide XDR and Amplified Secondary Antibody Detection Kits, these three-step incubation protocols enable maximum optimization of assay sensitivity.
