Neurobiological Markers of Cocaine Use Disorder

Cocaine use disorder is characterized by chronic cycles of abstinence and relapse; prevention and treatment of cocaine use disorder could be greatly advanced by a more comprehensive understanding of the neurobiological underpinnings. Alterations in the serotonin and glutamate neurotransmitter receptor systems in the brain are putative molecular mechanisms which may be involved in the propensity to relapse to cocaine use. We optimized and implemented a quantifiable and reproducible automated Simple Western assay in Wes™, to examine serotonin and glutamate receptor expression as well as protein:protein interactions from rodent brain samples.

Drug product characterization and target validation are critical steps in biopharmaceutical development. Both involve understanding the role of a certain biomolecule, such as protein, in a disease process. Traditionally, Western blots have been the default technology for confirming the presence or absence of a protein during these steps in drug development, but it suffers from a tedious and manual process that does not offer throughput or reproducibility. The Simple Western platform is the modern evolution of traditional immunoassay technologies and is uniquely suited to handle the rigorous needs of product characterization and drug target validation. Approaches to the Use of Limited Clinical Material for Oncology Biomarker Study and Characterisation

The need for biomarkers or surrogate biomarkers in cancer and other diseases is pressing. Ethical and other issues, however, often limit the amount of material available for bioassay. We therefore developed methodologies appropriate for limited sample sizes deriving from clinical respiratory disease and cancer research. We have characterized a novel approach to this issue using the ProteinSimple's Simple Western charge assay run on the NanoPro 1000 capillary-immunoassay instrument. We have developed assays encompassing clinical samples from Chronic Myeloid Leukaemia (CML-SC), Chronic Lymphocytic Leukaemia (CLL), Non-Small Cell Lung Cancer (NSCLC; tissue & plasma) and Endometrial Cancer. This and other work with Protein Simple has defined the utility of the Simple Western charge assay in detailed mapping of signaling cascades in model systems and cellular organelles. A range of studies across these disease areas are currently ongoing for validation of disease and/or treatment markers in biopsy material and plasma. This includes work designed to extend our knowledge of aberrant post-translational events in human primary disease tissue samples as compared to control material. Furthermore, we have developed protocols for cross-platform analysis of samples to expedite results validation from high-end MS proteomic analysis creating a platform for validation/verification on small samples of clinical material.

Biotherapeutic Development is Hard: ProteinSimple Delivers Tools to Simplify the Process

In order to bring an antibody or other biologic drug to market, each step of the process needs to be carefully monitored. Charge heterogeneity and apparent MW via SDS-PAGE are two of the most commonly assessed parameters. Ideally, similar methods should be used from the beginning of development (low expression, complex matrix) to late in production and QC (high concentration, pure material). This enables comparison of early development findings to product release data.

Capillary-based techniques such as iCE and CE-SDS are currently heavily used in downstream product development, but pose challenges for small scale screening and characterization during cell culture and formulation development. In this webinar, we will discuss the latest Simple Western product, Peggy, which combines capillary electrophoresis with an immunoassay to provide highly reproducible and fully automated analysis of monoclonal antibodies.

This sensitive instrument measures either size or charge in complex samples and provides critical charge heterogeneity, size, and product titer information without the need for sample purification.
